Brief introduction of IL17 family

The main effector of Th17 cells is IL-17. The study found that IL-17 is a T cell-derived cytokine that includes six member ligands (IL-17A~IL-17F) and five receptors (IL-17RA~IL-17RD and SEF). IL-17 is a proinflammatory cytokine produced primarily by activated T cells, and most of the immunomodulatory functions of IL-17 family cytokines have been reported, probably because of its induction of multiple immune signaling molecules. The most striking role is that IL-17 is involved in the induction and regulation of pro-inflammatory responses. IL-17 is usually associated with an allergic reaction. IL-17 induces a variety of cell types (fibroblasts, endothelial cells, epithelial cells, keratinocytes, and macrophages) to produce a variety of cytokines (eg, IL-6, G-CSF, GM-CSF, IL-1β, TGF). -β, TNF-α), chemokines (including IL-8, GRO-α and MCP-1) and prostaglandins (such as PGE2). The release of cytokines triggers a variety of effects, such as airway remodeling, a characteristic of the IL-17 effect. Increased expression of chemokines attracts other cells including neutrophils but not eosinophils. A subset of IL-17 to CD4+ T cells is referred to as helper T17 cells (Th17). Because of these effects, the IL-17 family has been closely associated with many immune/autoimmune related diseases including rheumatoid arthritis, asthma, lupus erythematosus, transplant rejection, anti-tumor immunity, and psoriasis.

IL-17A

The initial description of the IL-17 protein, now known as IL-17A, is a homodimer of two 136 Amino Acid chains, secreted by activated T cells, acting on stromal cells to induce inflammatory responses and hematopoietic active molecules. Today, IL-17 represents a structurally related family of cytokines that share a highly conserved C-terminal region but differ from each other's N-terminus and have different biological roles. Six members known in this family, from IL-17A to IL-17F, are secreted as homodimers.
Human IL-17A can cross-react to mouse cells.

IL-17B

IL-17B is two 161 amino acid polypeptide chains joined by disulfide bonds. T cells express IL-17B and stimulate the release of TNF-a and IL-1β by monocyte lineage cells.

IL-17D

IL-17D is a two 185 amino acid polypeptide chain linked by a disulfide bond. IL-17D has the ability to stimulate the production of IL-6, IL-8 and GM-CSF, and inhibits the hematopoiesis of myeloid progenitor cells in colony forming assays.

IL-17E

IL-17E is a two 145 amino acid polypeptide chain linked by a disulfide bond. IL-17E stimulates the secretion of IL-8 and induces the activation of the transcription factor NK-kB in IL-17BR receptor cells.

IL-17F

IL-17F stimulates proliferation and activation of T cells and PBMCs. IL-17F also regulates cartilage matrix reversal and inhibits angiogenesis.
