experimental method:
1. The gray matter tissues such as the cerebral cortex of neonatal rats were excised, and the isolated cell suspension was prepared by shredding and trypsin digestion, and the primary culture was carried out in the culture flask for about two weeks. Preparing a mixed glial cell primary culture;
2. The primary culture of glial cells was treated with shaking at 150r/min for 2h on a 37°C shaker.
3. Collect the cell suspension shaken in the culture flask and plant it in a plastic petri dish coated with no growth substrate component for short-term culture;
4. Discard floating cells and suspensions that are not attached. The cells that have been attached to the culture dish are microglia;
5. The obtained microglia are prepared into a cell suspension having a cell density of 2×10 5 /ml;
6. Plant the cell suspension in a culture flask or dish and continue the culture. At this time, 10% crude CFS or 25 U/ml pure CFS was added to the culture solution. Change the liquid twice a week;
