The physicist organization network reported on May 13 (Beijing time) that the substance named lomaiviticin A has been previously proven to kill human tumor cells in culture dishes, but its mechanism of action has not been revealed for more than a decade. . In a series of experiments, Yale University scientists Seth Herzo, Peter Reiser and colleagues found that this substance can kill by etching, cutting and eventually destroying the DNA of cancer cells and blocking their replication process. cancer cell.

Herzo said that DNA is the target of many anticancer drugs, and DNA strands that directly destroy cancer cells are one of the most effective ways, but few anticancer drugs can directly cut DNA. Lomaiviticin A, produced by deep-sea bacteria, has this ability, and it is expected to be an effective chemotherapeutic drug, and it can be used in small doses.

This substance contains two diazoxides (functional groups that determine the chemical nature of the substance) and is closely involved in the entire process of DNA cleavage while shearing the standard DNA structure. For ease of understanding, you can compare lomaiviticin A to a pair of scissors, and DNA is the two vines that you want to trim. Just like cutting two vines, when they interact with DNA, they can simultaneously cut the two strands of DNA at one point.

This deep-sea bacterium also produces two other related substances, lomaiviticin C and kinamycin C, but both of them contain only one diazepine and cannot complete DNA cleavage.

Herzo said: "Although we don't know what kind of interaction between lomaiviticin A and DNA, and what ultimately leads to DNA breaks, one thing is clear: it is destroying cancer. The substance has a powerful ability on the DNA of the cell."
